Speakers are usually mylar units and should protect it from entering unit and causing problems..Many cover same and might try a vacuum cleaner or perhaps a straw to remove...
 
As Dan said you should be able to vacuum the sand out. No real need to open the box. Do you use headphones? If so you might want to tape over the speaker grill. I have always used electrical tape to cover mine.
